Obiettivi formativi/Mission
The course aims to provide students with adequate information regarding: – gross anatomy of skin and microscopic confidence in its structure; – technical skills to study in vitro skin regeneration; – search in the literature to develop an idea of skin regeneration identifying strengths and weaknesses; – multiple roles of the skin in homeostasis including protection, regulation of body temperature, sensory reception, water balance, synthesis of vitamins and hormones, and absorption of materials.

Modalità di verifica dell'apprendimento/Assessment methods
The exam mark will be composed of two parts: a written test that will cover the material delivered in lectures and seminars (50% of the mark) and a project work in the form of a written essay that will cover the literature knowledge. The topic will be assigned by the lecturers and will be due on the same day of the exam. Written test: the written exam will take place in the computer room and will consist of 50 (25 for Anatomy and 25 for Physiology) closed questions. Multiple choice questions, true / false questions, completion type questions and matching type questions are the most frequent types of test questions. Written test evaluation criteria: True/ False Questions: 1 point for correct answer; -1 point for wrong answer; 0 points for not responding. Multiple choice questions: 1 point for correct answer; – 0.5 points for the wrong answer if the question allows you to choose from three options, -0.33 between four, -0.25 out of five, etc.; 0 points not responding. The penalty scores will be homogenously distributed for all tests. Rules for the written test – The test will be monitored: No Internet, No Second Screen, No outside help. You will be allowed 1 hour to complete all 2 sections (Anatomy and Physiology) of the test. Written essay evaluation criteria: Balanced fair and correct Synthesis and communicative skills Timely and pertinent Rules for the written essay – max 2 pages, single line spacing, character 12 + figures (optional) + bibliography and graphical abstract (mandatory)
Programma esteso/Content
ANATOMY. Gross structure of skin and underlying tissues, of the skin appendages and of the skin lines. Skin structure: layers of epidermis, the papillary dermis, the reticular dermis, hypodermis. Microscope-Learning 1: the skin and the hypodermis. Appendages of the skin: nails, hair and hair follicles, sebaceous glands, sweat glands. The skin throughout life. Disorders of the integumentary system. Microscope-Learning 2: appendages of the skin and examples of disorders. In vitro models to study skin tissue regeneration. PHYSIOLOGY Functions of the Integumentary System. The role of the skin in the protection from invasion by microorganisms, chemicals and other environmental factors and in preventing the body dehydration. Endocrine functions of the skin. The role of the skin in the body thermoregulation Sensory functions of the skin.
Risultati di apprendimento attesi/Intended learning objectives
The students will acquire comprehensive theoretical knowledge of the structure and role of skin in the human body physiology. They will develop a substantial set of skills, including the ability to critically analyse and discuss clinical and experimental themes related to skin structure and functions and the knowledge of their possible applications in other scientific fields. They will acquire the use of specific scientific and technical language improving their competence in the scientific communication.

Programma esteso/Content
Cellular physiology: electric activity of the cell; ionic signaling; elements of signal transduction. Skin as an organ, skin functions. Thermoregulation: receptors, mechanisms of signal transduction, neural paths, coordingation of central and peripheral thermoregulation. Sensation: types of sensation, receptors, mechanisms of signal transduction. Hair physiology: cellular organization, functions, integration with systemic physiological responses. Sweat physiology: types and cellular physiology of sweat glands; control of sweating; role and mechanisms in human health. The program may be subjected to modifications.

The students will learn the basis of mucosal immunity, the pathogenesis of inflammatory skin disorders, either autoimmune disorders or allergies, and the cellular and molecular mechanisms underlying wound healing. The students will also gain knowledge on the physiological microbiota of skin and on the principal pathogens associated with skin diseases

Obiettivi formativi/Mission
The course aims to: - provide students with adequate information about the skin as an ecosystem, the structure and function of the skin microbiota and factors affecting the microbial flora of the skin. - provide students with the basis for the understanding of the main microbial skin pathogens/host interaction mechanisms, microbial skin infections, microbial life in biofilms and their adverse effects on health. - provide students with the instruments for the application of some of the principal methods concerning bacterial and fungal cells cultivation and microscopic observation. - provide students with a comprehensive knowledge of skin immunity and of the pathological mechanisms underpinning the most common skin disorders. - provide students with the competences to address unmet clinical needs by suggesting experimental approaches that could be taken to identify new targets/approaches and to analyse their therapeutic potential.
Prerequisiti/Required background knowledge
Basic knowledge of microbial cell biology, genetic and metabolism that can be acquired by studying the textbook “Brock Biology of Microorganisms”. Basic knowledge of cell biology and immunology is mandatory and can be acquired by pre-course intensive tutoring. Anatomy, physiology and molecular biology are recommended.

The exam will be aimed at evaluating the knowledge and notions acquired during the lectures and selected seminars. The exam will be a written test composed of 4 open questions (answers of 150-200 words maximum) and 10 multiple choices and true/false questions (with one true answer). The questions will be aimed at evaluating knowledge and understanding of the topics addressed during the lecturers and the ability to discuss the topics critically and concisely by using specific scientific and technical language. At each open question a maximum score of 5 points will be assigned. For multiple choice and true/false questions, 1 point will be obtained for the correct answer, whereas wrong or blank answers will be evaluated 0 points (maximum score will be 30 points). Teams composed of 3-4 students/each will have to prepare a journal club presentation. A list of research manuscripts that falls into the topics discussed during skin pathology lectures will be provided. However, students might discuss additional original research articles, provided that the manuscript is approved by lecturer. The scientific paper will be discussed with all classmates – who will gain valuable insights into the different topics - and lecturer, who will evaluate the students work. Each student might be awarded with a maximum score of 3 points that will be added to the score of the written test.

Microbial life on skin: importance for human health and implications in skin diseases. Frontal lessons (Fracchia L.) - Brief introduction about skin as an ecosystem. Definition of Microbiota and Microbiome. Sites of microbial colonisation on the skin and influence of skin components and properties on microbial life. Overview of the principal methodologies to study the human microbiota. - Structure of the healthy human skin microbiota. Functions of the healthy human skin microbiota. Skin microorganisms or communities with and adverse or sickening effect on their hosts and relations with skin microbiota. - Microbial biofilms and skin infections. Prebiotics, probiotics, postbiotics and the skin microbiota in the cosmetic industry. The good and the bad: a close sight to some microorganisms belonging to skin microbiota and to those involved in skin diseases. Laboratory (Fracchia L.) - Theory and practice of microorganism cultivation on selective media, observation of relevant skin pathogens cultivated on agar plates, Gram- staining and observation of slides by optical microscopy. - Isolation and identification of microorganisms from skin surface. - How to cultivate a biofilm in vitro Skin immunity. Overview of the immunological anatomy of the epidermis and derma biology under steady state, wound healing and skin disorders. Discussion of research articles Porta C. At the end of the course, the student will acquire theoretical knowledge and understanding about the composition and the role of the physiological human skin microbiota as well as about the skin microorganisms or communities with adverse effect on human health and the main strategies to limit microbial invasion. The student will acquire the instruments for the application of the acquired knowledge to critically analyse and discuss clinical and experimental themes in the field of skin microbial diseases and for the application of some of the principal methods for cultivation and observation of microorganisms. The student will gain independence of judgment on the different topics addressed during the course, knowledge of their possible applications and repercussions in other scientific fields and skills regarding the verbal and written communication by the use of specific scientific and technical language. The student will acquire theoretical knowledge and understanding of cellular and molecular mechanisms underpinning cutaneous immunity, pathogenesis of inflammatory and infectious skin disorders. The student will acquire the specific scientific terminology and application of the acquired knowledge to critically analyse and discuss clinical and experimental data in the field of skin immunity and disorders. Finally, the student will gain competence to conceptually design small research projects aimed to: 1) discover new targets, 2) study the therapeutic potential of a given target 3) evaluate new approaches to target key pathogenic events.

The course aims to: - provide students with adequate information about the skin as an ecosystem, the structure and function of the skin microbiota and factors affecting the microbial flora of the skin. - provide students with the basis for the understanding of the main microbial skin pathogens/host interaction mechanisms, microbial skin infections, microbial life in biofilms and their adverse effects on health. - provide students with the instruments for the application of some of the principal methods concerning bacterial and fungal cells cultivation and microscopic observation.

Basic knowledge of microbial cell biology, genetic and metabolism

Modalità di verifica dell'apprendimento/Assessment methods
The exam will be aimed at evaluating the knowledge and notions acquired during the lectures and selected seminars. The exam will be a written test composed of 4 open questions (answers of 150-200 words maximum) and 10 multiple choices and true/false questions (with one true answer). The questions will be aimed at evaluating knowledge and understanding of the topics addressed during the lecturers and the ability to discuss the topics critically and concisely by using specific scientific and technical language. At each open question a maximum score of 5 points will be assigned. For multiple choice and true/false questions, 1 point will be obtained for the correct answer, whereas wrong or blank answers will be evaluated 0 points (maximum score will be 30 points). Teams composed of 3-4 students/each will have to prepare a journal club presentation. A list of research manuscripts that falls into the topics discussed during skin pathology lectures will be provided. However, students might discuss additional original research articles, provided that the manuscript is approved by lecturer. The scientific paper will be discussed with all classmates – who will gain valuable insights into the different topics - and lecturer, who will evaluate the students work. Each student might be awarded with a maximum score of 3 points that will be added to the score of the written test.
Programma esteso/Content
Frontal lessons: Microbial life on skin: importance for human health and implications in skin diseases. - Brief introduction about skin as an ecosystem. Definition of Microbiota and Microbiome. Sites of microbial colonisation on the skin and influence of skin components and properties on microbial life. Overview of the principal methodologies to study the human microbiota. - Structure of the healthy human skin microbiota. Functions of the healthy human skin microbiota. Skin microorganisms or communities with and adverse or sickening effect on their hosts and relations with skin microbiota. - Microbial biofilms and skin infections. Prebiotics, probiotics, postbiotics and the skin microbiota in the cosmetic industry. Laboratory: The good and the bad: a close sight to some microorganisms belonging to skin microbiota and to those involved in skin diseases. - Theory and practice of microorganism cultivation on selective media, observation of relevant skin pathogens cultivated on agar plates, Gram- staining and observation of slides by optical microscopy. - Isolation and identification of microorganisms from skin surface. - How to cultivate a biofilm in vitro
Risultati di apprendimento attesi/Intended learning objectives
At the end of the course, the student will acquire theoretical knowledge and understanding about the composition and the role of the physiological human skin microbiota as well as about the skin microorganisms or communities with adverse effect on human health and the main strategies to limit microbial invasion. The student will acquire the instruments for the application of the acquired knowledge to critically analyse and discuss clinical and experimental themes in the field of skin microbial diseases and for the application of some of the principal methods for cultivation and observation of microorganisms. The student will gain independence of judgment on the different topics addressed during the course, knowledge of their possible applications and repercussions in other scientific fields and skills regarding the verbal and written communication by the use of specific scientific and technical language.

Working with data: use of the R software for statistical analysis and graphics. Epidemiology: epidemiological methods in public health; epidemiological study designs (clinical trials, cohort studies, case control, transversal and descriptive studies); sources of uncertainty (role of chance, bias and confounding). Systematic reviews and meta-analysis: protocol writing, meta-synthesis of qualitative data, meta-analysis and risk of bias assessment.

An online examination accounting for the 75% of the total grade to be carried out on the exam day and will be delivered through DIR. This in turn will be subdivided in: – a set of multiple-choice/answer questions (2/3 of the points) in the theoretical contents of the course. – a set of exercises involving statistical analyses/procedures (1/3 of the points). A written essay to be done at home assigned by the lecturer of the module entitled “Systematic review and meta-analysis” (25% of the final mark). Each student will be asked to write the protocol of a systematic review, followed by a meta-analysis, on a topic of his/her choice. The essay must be delivered approximately 15 days before the exam day (the exact deadline for submitting the essay will be communicated during the course).

Statistics: Basic elements of descriptive statistics. Single variable summary statistics (mean, and other statistical indicators) and graphics (histograms and boxplots). Double variable and regression models. Probability and discrete random variables Continuous random variables. The gaussian. Area under the normal distribution. Standardization. Confidence intervals. The Student t-distribution Statistical hypothesis testing. The t-test. 2 sample t-test.The p-value. One sided and two sided alternative hypothesis. Chi square test ANOVA Introduction to R. Import data from excel to R. Plots in base R and using the R package ggplot2. Data simulation. Statistics with R. Epidemiology: Definition of epidemiology. Measures of occurrence and association. Main Epidemiological studies: clinical trials, cohort studies, case-control studies, cross-sectional studies, descriptive studies. Sources of uncertainty in epidemiology: the role of chance, bias and confounding. Systematic review and meta-analysis: refine research question, protocol writing and registration, meta-synthesis of qualitative data, meta-analysis techniques, the risk of bias assessment. In addition to the theoretical knowledge, students will gain practical experience by designing a systematic review and meta-analysis, including writing a protocol on a pre-assigned topic.

Obiettivi formativi/Mission
People involved in the development of drugs have really diverse educational backgrounds and cooperate with their complementary knowledge and skills to the final goal of R&D. The course aims at conveying a general knowledge on the drug discovery and development process so that students from different backgrounds may be able to appreciate the meaning and importance of the different phases of R&D, become familiar with key terminology, understand complexity and identify critical factors and bottlenecks that impact the discovery of medicines. Finally students may also be able to critically understand the process that allowed a new medicine to be approved and, if given the opportunity, communicate/cooperate across the different fields of the R&D process.

Metodi didattici/Teaching methods
Teaching is given in the form of: i) classroom lectures; ii) practicals including literature and data searches; iii) discussions of case studies. Additionally, the course is enriched by 3 h seminars by the CEO of the start up company ChemiCare (Dr. Beatrice Riva) who will provide additional and personal insights into R&D process and drug development with particular reference to GXP compliance and ICH (international Council for Harmonization of Technical Requirements for Pharmaceuticals for Human Use) guidelines.

The students, in small groups, will be required to search and retrieve data on a recently developed drug for skin disorders and critically evaluate either preclinical and/or clinical aspects of the R&D process in a oral presentation.

The different phases of R&D and the different professional profiles involved in R&D; early and key decisions in R&D; preclinical in vitro and in vivo models in drug discovery; general principles of drug screening and assay development for drug screening; clinical trials; critical discussion of R&D process of drugs for skin disorders recently licensed; insights from being a CEO of a start up company involved in R&D (3 h seminar by Dr. Beatrice Riva): mindset, challenges, personal skills, business and market opportunity.

Upon course completion, the student should be able to: i) understand the significance and relevance of the different and distinct phases of the R&D process; ii) recognize key decision points in R&D; iii) carry out searches aimed at retrieving information relevant to the development of a drug; iv) review and critically evaluate preclinical and clinical studies with a general understanding of their aim based on medical needs, of chosen procedures, of results, conclusions and relevance.

The course aims to provide students with the basic principles of the formulation, illustrating in particular the physical-chemical characteristics of the excipients used for the preparation of dosage forms for topical application. An overview of skin care, sun care and decorative products will also be presented. Moreover, the course aims to provide the students with adequate information regarding manufacturing techniques of topical dosage forms; preparation of makeup and skincare products.

The written test will consist of multiple choice (20 questions for each 4-hour block): true/false type questions are the most frequent types of questions. 1 point for the correct answer, 0 points for wrong or blank answers. The exam relating to the cosmetic formulation will take place during the laboratory in the form of multiple choice questions (relating to the lectures) and the performance of the students during the practical part of the course. Both tests aim to verify the knowledge and ability of the students to apply in an independent way the knowledge acquired. Moreover, the communication skills of the students will be evaluated, particularly the use of specific language for the technological discipline. During the practical laboratory, the ability to apply the knowledge acquired will be evaluated, considering the accurate presentation of the topical preparation. The evaluation will consider the learning abilities of the students, in particular the ability to deal with formulative challenges in the field of topical preparations. The exam will take place during the official dates, and the grade will be expressed in thirtieths, as the average mark obtained for each part of the course.

Obiettivi formativi/Mission
Il corso intende promuovere la conoscenza delle basi generali delle principali patologie cutanee (a genesi infettiva, autoimmunitaria, neoplastica), con particolare attenzione al trattamento delle varie condizioni. Inoltre si intende porre l'attenzione al ruolo del microbiota cutaneo nelle varie patologie (es atopia, psoriasi, dermatiti infettive). Verranno trattate le basi del danno cutaneo UV-mediato e ella fotoprotezione. Infine verrà presentata un'introduzione alla cosmetologia ed alla medicina rigenerativa in dermatologia.
The course aims to allow students to understand the general principles of the main skin diseases, thus including either infectious, immune-inflammatory, or neoplastic disorders (as listed in the program). A special attention is paid to the current dermatological treatments of the mentioned diseases. The role of cutaneous microbiome in different dermatosis (i.e. atopy, psoriasis, cutaneous infections) is also discussed. Ci attendiamo che alla fine del corso l* student* abbia una discreta conoscenza delle principali malattie cutanee, con particolare riguardo alla loro patogenesi e terapia. Inoltre l* student* dovrà essere in grado di capire correttamente e discutere articoli scientifici di ambito dermatologico. In considerazione di ciò, l* student* sarà in possesso di un background scientifico sufficiente per poter proporre/sviluppare nuovi possibili terapie per le malattie cutanee.
At the end of the course, the student is expected to thoroughly know the general principles of the main skin disorders, with special regard to their pathogenesis and current treatments. It is also expected the student to be able to understand and criticize articles from the dermatology literature. On these bases, we aim to provide the student the suitable scientific background in order to propose other possible pathway/drugs able to treat some skin diseases.

The students are introduced to the main functional groups and heterocycles, their electronic properties and reactivity. Selected examples of synthetic routes to pharmaceutical and cosmetic compounds are illustrated. The theoretical knowledge is applied in the laboratory by setting up a set of reactions and purifying the corresponding products. The synthesis leads to the preparation of a dermatological API that will be then formulated during the formulation laboratory in the second semester.

The exam mark will be the average mark obtained for each module: Theory: The students will be divided in groups and each group will have to present two projects in the form of a Powerpoint presentation: one relating to an organic chemistry topic and one relating to the analysis of a synthetic strategy of a product for pharmaceutical and/or cosmetic use. Each project will be assigned a maximum score of 15 points Practice: (i) The written test covers the material delivered in lectures and seminars and is made of multiple-choice questions (20) and an exercise (6). Maximum score: 26 For multiple choice questions, 1 point will be obtained for the correct answer, whereas wrong or blank answers will be evaluated 0 points. The laboratory evaluation considers the commitment, the performance of the student in the laboratory, and the lab notebook writing. Maximum score: 6.

The course will cover the key aspects of quality assurance of cosmetic and dermatological products: quality assessment during product development (Quality by Design approach); implementation of stability tests of active pharmaceutical ingredients (API) and finished products according to international regulatory guidelines (ICH); principles of qualitative and quantitative analysis for determining the identity and purity of APIs and their potency in the finished products.

Students will be provided with the slides projected during lessons. Original scientific articles, useful for understanding the topics covered during the course, will also be provided.

Quality control (QC) of pharmaceuticals involves a series of rigorous processes and testing procedures to ensure that products meet specified standards and regulatory requirements. By attending the course, the student will be able to manage the theoretical and practical aspects of quality control in pharmaceutical products from the raw materials to the finished product and the most common analytical techniques involved.

At the end of the course, which also includes laboratory activities, the student will have knowledge of the key aspects of quality assurance of cosmetic and dermatological products. They will also have knowledge of the analytical procedures used to determine the purity of the active pharmaceutical ingredients and their potency in the corresponding formulations. Furthermore, the student will be able to calculate, interpret and evaluate the analytical data obtained.

Approach to natural active pronciples for cosmetic and dermatological use considering the advantages and disadvantages of natural compounds application as pure entities or as a phytocomplex. Classes of primary and secondary metabolites with their chemical and physiscal characteristics and different applications. From classic to modern techniques of extraction and evaporation applied to vegetable material. Classic and modern techniques applied for isolation and analysis of natural compounds.

Obiettivi formativi/Mission
Students will learn what a primary and secondary metabolite are and why plants produce them. They will learn and recognize classifications of natural compounds and the possibilities of interactions with our biology. They will understand important functional groups leading to precise biological effects and new biological aspects. Students will also learn different purification technologies that allow to obtain pure compounds, how to choose the best one, and how to scale up a method from the laboratory to an industrial setting. After that, students will be able to choose the best technique for the isolation of a pure active ingredient or the analysis required for the quality control and/or stability.

Organic chemistry, biology and pharmacology.

Lectures with computer and video supporting: projection of powerpoint slides. Interactive lectures with discussion. Vegetable samples, extracts, different oils, essential oils and pure compounds will be shown to be touched, smelled and compared to demonstrate practically the physical, chemical and olfactory differences.

A written exam evaluating the knowledge, notions and processes acquired during the lectures. The test includes: a) 3 open questions with answers of 100-150 words maximum, 6 points each. 60% of the final grade. b) 10 multiple choice questions with one true answer, 3 points each. Correct answer gives 1.2 point. All other possibilities is 0 point. There will be no negative grades. 40% of the final grade. The overall grade will be the sum of the two parts.

What is a natural product? Phytocomplex and the entourage effect. Primary and secondary metabolites: definition, roles and classification. Pure natural compound and phytocomplex: advantage and disadvantage. Primary metabolites: oils, butters, waxes. Stability and characteristics. Classification and characteristics of secondary metabolites: – alkaloids: biosynthesis and isolation, a matter of basicity – isoprenoids: from essential oil composition to chemestetic properties of functional groups, how to recognize, how to apply them (TRPV3, TRPA1, TRPV1, hTAS2R) – meroterpenoids and TRPV1 – phenyl propanoids and polyketides Drying process. Solvents: differences in polarity and application. NADES. Extraction process: classic techniques as maceration, percolation, infusion and decoction, distillation, cold expression. Modern techniques: ASE, MWE, sonication, SFE, electroporation, enzymatic extraction. Rotavapor, wiped film distillation, sray drying. Isolation techniques with stationary phases: differences and how to choose the appropriate one. HPLC, FLASH chromatography and GC. Definition and application. Other processes: distillation, liquid-liquid partition, avoiding of fatty acids and waxes. Every aspect of the program is supported by examples of natural compounds and ther application in the dermatological/cosmetic field.

Students should be able to recognize different classes of primary and secondary metabolites and their physical, chemical characteristics and behaviors. They should also be able to plan extraction and isolation strategies. All the principles and techniques learnt during the course will be put in practice during the laboratory included in the course.

This course is a writing laboratory where students will immediately apply what they are learning. The lab shall work as a little newsroom where teamwork and constant improvement are an effective part of the journey. Writing and producing material will be solo work, but the entire class shall help each other for the better result.
Modalità di verifica dell'apprendimento/Assessment methods
I shall evaluate how the article is written, if it is suitable and understandable for a general public (therefore strong jargon shall be avoid as much as possible), if it complies the Seo rules, if it is delivered on time as well as if it is paired with the proper photos. The scientific mentor shall evaluate the scientific content considering its originality, the strong basis on which the assumptions are made, the reliability of the sources and the observations made by the students. The final mark will be expressed in 30/30 with the possibility of the laude if the product will be consistent, wide and curated in all details.
Programma esteso/Content
In the first part I will explain how to write an article for the online student magazine aimed at a general public. The goal is to produce at least one product per student to be published on https://www.emotion-master.eu/emotion-student-magazine/ The product shall be an article with photos and other possible elements such as video, audio, infographics. We dive into how to choose a topic of interest, how to choose a title and how to deliver the message. Then we will focus on the SEO writing for a better positioning of the article.

The student shall write at least a scientific article with photos. Photos can be taken personally, produced with AI or taken from website with free common licence.

The course provides the students with the main information about ADMET of dermatological drugs. The course also provides the students with the principles for the discovery and optimization of a bioactive compound, from hit to lead compounds to drug candidates. The design of prodrugs and soft drugs for topical use is covered. Case studies on dermatological drugs are presented from both a pharmacological and medicinal chemistry perspectives.

Obiettivi formativi/Mission
1. At the end of the course the student will know and understand the basic concepts of pharmacology and medicinal chemistry, in particular the route followed by the drug in the organism, from its administration to its excretion and how it interacts with its molecular target, focusing mainly on oral and topical administration. Moreover, the path of the drug in the pharmaceutical industry, from drug discovery to drug development through drug design will be addressed. 2. The student will apply the acquired knowledge and understanding in the recognition of the chemical structures, in the discussion of the chemical and chemical-physical properties, of the mechanism of action, of the structure-activity relationship and of the metabolic fate, together with toxicological aspects. 3. The student will acquire the ability to analyze texts and papers in the pharmacology and medicinal chemistry literature. 4. The student will learn to communicate about his/her understanding, to describe, even in an original way, a topic described during lessons, to adequately answer to questions, critiques and suggestions. He/she will also learn how to present a drug to the public using the information searched in the scientific literature and in databases. 5. Modalità di verifica dell'apprendimento/Assessment methods
Written test. It covers the material delivered in lectures. Multiple choice, true/false and completion type questions are the most frequent types of questions. 1 or 2 points for the correct answer, 0 points for wrong or blank answers. Oral presentation. Students will be given a dermatological approved small molecule. Working individually, they will need to cover the main medchem aspects (metabolism, synthesis, intermolecular interactions, discovery and optimization, stereochemistry, LogP, LogD, etc.) and pharmacology aspects (mechanism of action, pharmacokinetics, therapeutic need, competitors, and place in therapy). The Case Study will be orally presented by the students with the support of slides in a 10’ presentation. The total mark is the average between the two marks.

Absorption. Mechanisms of absorption with a focus on skin. Phase I and phase II metabolism with a focus on skin and liver metabolism. Distribution and elimination of drugs. Principles of pharmacodynamic. Drug-target intermolecular interactions and SAR studies. Role of stereochemistry in the interaction with the binding site. Receptors and enzymes as drug targets. Drug discovery. Hit compounds and lead compounds. How to recognize drug-like molecules. LogP and logD. Lipinski rule and beyond. Strategies for lead optimization. Prodrugs and soft drugs. Case studies of dermatological drugs.

At the end of the course, the student will know the principles of general pharmacology and medicinal chemistry. He/she will be able to apply these principles to a specific bioactive molecule and to evaluate its chemical, metabolic and drug-likeness properties. The student will also know the role of medicinal chemistry and pharmacology in drug discovery and in drug design and the strategies that can be used in order to improve the pharmacodynamic and pharmacokinetic properties of a dermatological drug.

Contenuti/Content Summary
Il corso coprirà diverse sezioni chiave e contenuti correlati per fornire alle studentesse e agli studenti una comprensione approfondita della scienza delle proteine e delle sue applicazioni. Inizierà con una sezione introduttiva di livellamento in cui saranno discussi i principi e le nozioni di base sulle proteine e sugli enzimi, comprese le loro proprietà e funzioni. Successivamente, il corso presenterà e discuterà i principi di base del drug design e dell'ingegneria delle proteine su base razionale. Saranno esplorati anche i metodi fisico-chimici per lo studio delle interazioni molecolari proteina-proteina e proteina-ligando. Inoltre, ci sarà un laboratorio focalizzato sui danni al DNA e la riparazione, oltre che sugli antiossidanti a base proteica. Il corso includerà seminari tenuti da esperte ed esperti di aziende impegnate nel campo della scienza delle proteine applicata alla cosmesi. Inoltre, il corso si propone di introdurre le studentesse e gli studenti ai principi principali delle simulazioni computerizzate di entità chimiche, inclusi composti e proteine. Attraverso queste sezioni integrate, le studentesse e gli studenti acquisiranno sia conoscenze teoriche sia competenze pratiche essenziali per simulare correttamente le strutture chimico-biologiche e comprendere le loro interazioni e applicazioni in vari campi.
The course will cover several key sections and related contents to provide students with a comprehensive understanding of protein science and its applications. It will begin with an introductory leveling section where the basic principles and notions of proteins and enzymes, including their properties and functions, will be discussed. Following this, the course will present and discuss the basic principles of drug design and protein engineering on a rational basis. The physical-chemical methods for the study of protein-protein and protein-ligand molecular interactions will also be explored. Additionally, there will be a workshop focusing on DNA damage and repair, as well as protein-based antioxidants. The course will include seminars led by experts from companies engaged in the field of protein science applied to cosmetics. Moreover, the course aims to introduce students to the main principles of computer simulation of chemical entities, including compounds and proteins. Through these integrated sections, students will gain both theoretical knowledge and practical skills essential for simulating chemical-biological structures and understanding their interactions and applications in various fields.

Obiettivi formativi/Mission
1. Conoscenze: L'obiettivo del corso è quello di migliorare la conoscenza delle studentesse e degli studenti sulla scienza delle proteine, con un particolare focus sulla relazione struttura-attività nelle macromolecole biologiche e sull'approccio strutturale per la progettazione razionale di potenziali molecole ligandi proteici con potenziale terapeutico. Dopo una fase iniziale di consolidamento sui concetti fondamentali, il corso procederà a contestualizzare i principi base della scienza delle proteine nel campo cosmetico, con un percorso didattico focalizzato su alcuni temi selezionati legati alla cosmesi, inclusi gli agenti antiossidanti e la riparazione dei danni al DNA. Inoltre, al termine del corso, le studentesse e gli studenti dovranno conoscere i principali metodi attualmente utilizzati nella simulazione di sistemi di interesse chimico-biologico. In particolare, dovranno essere consapevoli delle principali tecniche computazionali utili sia per il calcolo delle energie e delle geometrie molecolari, sia per l'analisi conformazionale di piccole molecole (farmaci, ligandi endogeni) e macromolecole (proteine e acidi nucleici). 2. Capacità di applicare le conoscenze: Le studentesse e gli studenti dovranno essere in grado di identificare la procedura più appropriata per risolvere vari problemi chimico-biologici proposti durante il corso. Dovranno inoltre saper utilizzare alcuni software di modellizzazione di base per prevedere la struttura tridimensionale delle proteine e l'associazione farmaco-recettore. Questo implica non solo la comprensione degli aspetti teorici, ma anche l'applicazione delle competenze pratiche per eseguire simulazioni e analizzare i risultati in modo efficace.
1. Knowledge: The objective of the course is to enhance the students' knowledge of protein science, with a particular focus on the structure-activity relationship in biological macromolecules and the structural approach for the rational design of potential protein ligand molecules with therapeutic potential. Following an initial phase of consolidation on the fundamental concepts, the course will proceed to contextualize the basic principles of protein science in the cosmetic field, with a didactic path focused on select cosmetic-related topics, including antioxidant agents and DNA damage repair. Additionally, by the end of the course, students will have to know the main methods currently used in the simulation of systems of chemical-biological interest. They must be aware of the main computational techniques useful both for the calculation of energies and molecular geometries, and for the conformational analysis of small molecules (drugs, endogenous ligands) and macromolecules (proteins and nucleic acids). 2. Ability to apply knowledge: Students must be able to identify the most appropriate procedure to solve various chemical-biological problems proposed during the course. They should also be proficient in using basic modeling software to predict the three-dimensional structure of proteins and the drug-receptor association. This involves not only understanding the theoretical aspects but also applying practical skills to perform simulations and analyze results effectively.

Le lezioni si svolgeranno ex-cathedra con il supporto di diapositive e materiale video, che saranno forniti alle studentesse e agli studenti all'inizio delle lezioni. L'aula sarà utilizzata per l'esecuzione di esercizi e quiz, nonché per la discussione dei dati presentati nella letteratura scientifica. Saranno introdotti argomenti generali e specifici, e saranno forniti concetti di base per facilitare la comprensione di ciascun tema. Inoltre, le studentesse e gli studenti potranno utilizzare i libri di testo consigliati per approfondire gli argomenti trattati in aula. Gli argomenti trattati in classe saranno anche oggetto di esercizi e simulazioni attraverso l'uso di programmi complessi di modellizzazione molecolare, permettendo alle studentesse e agli studenti di applicare le loro conoscenze teoriche in un contesto pratico.
Lessons will be conducted ex-cathedra with the support of slides and video material, which will be provided to students at the beginning of the lessons. The classroom setting will be utilized for the execution of problems and quizzes, as well as for discussions on the data presented in the literature. General and specific topics will be introduced, and basic concepts will be provided to facilitate the understanding of each issue. Additionally, students may utilize the recommended textbooks to further explore the topics covered in the classroom. The topics covered in class will also be the subject of exercises and simulations through the use of complex molecular modeling programs, allowing students to apply their theoretical knowledge in a practical context.

Modalità di verifica dell'apprendimento/Assessment methods
La valutazione finale consiste in un test con domande a risposta multipla sulla parte teorica del modulo delle proteine e del modulo di modellizzazione molecolare, che ha un peso totale di 4/5 del voto finale. Inoltre, le studentesse e gli studenti devono consegnare un breve saggio sulla parte di modellizzazione il giorno dell'esame, che ha un peso di 1/5 della valutazione finale.
The final assessment consists of a multiple-choice test covering the theoretical parts of the protein module and the molecular modeling module, which accounts for 4/5 of the final grade. Additionally, students are required to submit a brief assay on the modeling part on the day of the exam, which accounts for 1/5 of the final grade.

Si prevede che le studentesse e gli studenti saranno in grado di argomentare e partecipare a discussioni sui principi fondamentali della reattività delle macromolecole biologiche e sulla relazione struttura-funzione delle proteine. Inoltre, le studentesse e gli studenti dovranno dimostrare una comprensione approfondita del drug design basato sulla struttura. Al termine del corso, le studentesse e gli studenti avranno appreso gli aspetti fondamentali necessari per simulare correttamente le strutture chimico-biologiche. Saranno in grado di applicare questa conoscenza a problemi pratici, dimostrando la loro capacità di utilizzare efficacemente tecniche computazionali e software di modellizzazione.
It is expected that the students will be able to argue and engage in discussions on the fundamental principles of the reactivity of biological macromolecules and the structure-function relationship of proteins. Additionally, students should demonstrate a comprehensive understanding of structure-based drug design. By the end of the course, students will have learned the fundamental aspects necessary to correctly simulate chemical-biological structures. They will be able to apply this knowledge to practical problems, showcasing their ability to utilize computational techniques and modeling software effectively.

It is expected that the students will be able to argue and engage discussion on the fundamental principles of the reactivity of biological macromolecules and the structure-function relationship of proteins. Additionally, the student should demonstrate a comprehensive understanding of structure-based drug design.

Obiettivi formativi/Mission
1. Conoscenze. Al termine del corso, le studentesse e gli studenti dovranno conoscere i principali metodi attualmente utilizzati nella simulazione di sistemi di interesse chimico-biologico. In particolare, dovranno essere consapevoli delle principali tecniche computazionali utili sia per il calcolo delle energie e delle geometrie molecolari, sia per l'analisi conformazionale di piccole molecole (farmaci, ligandi endogeni) e macromolecole (proteine e acidi nucleici). 2. Capacità di applicare le conoscenze. Le studentesse e gli studenti dovranno essere in grado di identificare la procedura più appropriata per risolvere alcuni problemi chimico-biologici che verranno proposti durante il corso. Dovranno inoltre saper utilizzare alcuni software di modellizzazione di base per prevedere la struttura tridimensionale delle proteine e l'associazione farmaco-recettore.
1. Knowledge. At the end of the course students will have to know the main methods currently used in the simulation of systems of chemical-biological interest. In particular, they must be aware of the main computational techniques useful both for the calculation of energies and molecular geometries, and for the conformational analysis of small (drugs, endogenous ligands) and macromolecules (proteins and nucleic acids). 2. Ability to apply knowledge. The student must be able to identify the most appropriate procedure to solve some chemical-biological problems that will be proposed during the course. He must also be able to use some basic modeling software to predict the three-dimensional protein structure and the drug-receptor association.

Conoscenze di base in chimica generale, chimica organica, fisica e biochimica.
Basic knowledge of general chemistry, organic chemistry, physics and biochemistry.
Metodi didattici/Teaching methods
Il corso si svolge con il supporto di diapositive, che vengono fornite alle studentesse e agli studenti all'inizio delle lezioni. Saranno introdotti argomenti generali e specifici, fornendo i concetti di base per la comprensione di ciascun tema, e saranno tenute discussioni con le studentesse e gli studenti. Gli argomenti trattati in classe saranno oggetto di esercizi/simulazioni attraverso l'uso di programmi complessi di modellizzazione molecolare.
The course is given with the support of slides, which are given to the students at the beginning of the lessons. General and specific topics will be introduced, basic concepts will be provided for the understanding of each issue, and discussions with the students will be held. The topics covered in class will be the subject of exercises/simulations through the use of complex molecular modeling programs.

Modalità di verifica dell'apprendimento/Assessment methods
L'esame si compone di due parti (a) e (b), ciascuna parte concorre al voto finale per il 50%: (a) un questionario a risposta multipla di 30 domande che riguarderà le lezioni, i seminari e il materiale di lettura. Si otterrà un punto per ogni risposta corretta, mentre le risposte errate o nulle saranno valutate 0 punti; l’esame sarà cartaceo; (b) un elaborato che valuterà la capacità degli studenti di applicare le conoscenze generali fornite nei seminari ai loro domini regionali. In breve, agli studenti verrà chiesto di descrivere il quadro normativo nel proprio Paese relativo ai cosmetici, ai dispositivi medici o ai medicinali seguendo uno schema fornito dal docente. Ogni studente può adattare il contenuto dell'elaborato alla situazione specifica del proprio Paese. Gli studenti dei paesi extra-UE dovrebbero descrivere il processo che porta all'approvazione dei medicinali, le caratteristiche della loro agenzia regolatoria o le normative che regolano l'ingresso dei cosmetici o dei dispositivi medici sul mercato. Gli studenti europei dovrebbero descrivere come vengono valutati i medicinali nel proprio paese dopo l'approvazione dell'EMA, come rimborsati dai sistemi sanitari, dalle assicurazioni o dai privati. Se lo desiderano, possono anche scegliere un determinato prodotto cosmetico o un dispositivo medico e descrivere le peculiarità di quel prodotto rispetto all'intero regolamento cosmetico o dei dispositivi, ma il documento non deve riguardare il regolamento europeo generale. La valutazione verrà effettuata sulla qualità delle informazioni, sulla bibliografia, sulla capacità di rispondere allo schema fornito e sul risultato del Turnitin. È richiesto il report Turnitin. Verrà fatta la media dei due voti.
The exam is composed of two parts, each worth 50% of the final mark: (a) 30 multiple-choice questions that will cover lectures, seminars and reading material. One point will be obtained for the correct answer, whereas wrong or blank answers will be evaluated 0 points; the exam will be on paper; (b) a paper that will evaluate the students’ ability to apply the general knowledge shared at lectures and seminars in their own regional domains. Utilizing a specific format, students will briefly describe the regulatory framework in their own country related to cosmetics, medical devices or medicinal products. Each student may adapt the content of the paper to the specific situation of the country. Partner country extra-UE students should describe the process that leads to medicinal product approval, the characteristics of their regulatory agency and framework, or the regulations that govern the entrance of cosmetics or medical devices on the market. European students should describe how medicinal products are evaluated in their own country after EMA approval, as reimbursed by health systems, by insurances or by private citizens. If they wish, they may also choose a particular cosmetic product or medical device and describe the peculiarities of that product compared to the entire cosmetic or medical devices regulation, but the paper should not cover the general European cosmetic or medical devices regulation. Evaluation will be done on quality of the information, on references, on ability to answer at scheme provided, and on Turnitin report. Turnitin report is mandatory. The two scores will be averaged.

Obiettivi formativi/Mission
Il corso fornisce agli studenti una comprensione delle dinamiche dei mercati delle tecnologie sanitarie dal punto di vista delle Autorità di regolamentazione, delle Agenzie di valutazione delle tecnologie sanitarie, dei pagatori e dell'industria. Dopo aver completato con successo il corso, ci si aspetta che comprendano meglio quali sono le esigenze del mercato/dei pagatori, affrontando così le traiettorie di ricerca e sviluppo guardando a quando le tecnologie verranno introdotte sul mercato.
This course provides students with an understanding of the dynamics of the health technologies markets from perspectives of Regulatory Authorities, Health Technology Assessment Agencies, Payers and the Industry. After a successful completion of the course, students are expected to better understand which are the market / payers requirements, thus addressing the research and development trajectories looking at the future launch into the market.
